Modulation of ATP-Sensitive Potassium Channels in Rabbit Ventricular Myocytes by Endogenous Modulators

Abstract
Using patch-clamp techniques, we examined the effects of various endogenous modulators on k_(ATP) channel activity in single ventricular myocytes of the rabbit. Such modulators include ADP, stimulation of adenosine receptors, acidosis, and lactate, all of which are known to be generated during myocardial ischemia. They reduced the sensitivity of the channel to ATP. On the other hand, intracellular sulfhydryl oxidation caused an inhibition of the k_(ATP) channel. From our results, it could be suggested that the k_(ATP) channel activity may be modulated by various metabolic alterations that occur during myocardial is chemia. Metabolisminduced changes in the sulfhydryl redox can also modulate k_(ATP) channel activity.
